A 25th week trial (from 7th Jan. 1971-1st July. 1971. ) was conducted in order to compare the effect of different additives to the basic culture media KC (Knudson C), KM (Kano´s medium) and AMS (Murashige & Skoog) ect, on multiplication and shoot formation of protocorms obtained in meristem culture (solid cuture) of Cymbidium Rosanna $quot;pinkie.$quot;
The results obtained are given below:
1. For the growth of shoot in the early period (during first 5 weeks) the KC medium was found to be good. But for the growth of shoot portion during the middle and late periods (5th-15th week and 15th-25th week) the KM medium was found to be good.
2. For the growth of shoot portion during the middle and late periods (5th-15th week and 15th-25th week) the addition of organic substances viz; peptone (2㎎/ℓ), coconut milk(10%), banana juice (10%) to each of culture media KC, KM and MS showed good results.
Specially peptone showed hasting effect.
3. For the increase in fresh body weight of shoot portion the addition of peptone and coconut milk was proved to be good and more vigorous plant bodies were formed.
4. For the formation and growth of root portion the addition of banana juice was useful. Specially banana juice was more effective.
5. The addition of coconut milk to each of KC, KM and MS culture media was rot found useful because it tended to delay the formation and growth of root portion.
6. The addition of peptone was found to hasten the protocorm formation which is, useful for raising next culture.
7. The addition of peptone hastened chlorophyll formation.
8. The addition of banana juice and coconut milk to each of KC and KM culture media resulted in browning of cut surface of protocorm along with the portion of culture media just adjascent to it while showing no appreciable effect on growth.
9. In case of all the combinations of additives and MS culture media the cut surface of protocorm turned brown while the growth was slightly effected during early period (5 weeks) but showed normalization during later period (15-25 weeks).
10. According to the above experiment the best combinations of culture media and additives for growth are banana juice(KC and KM) and peptone (in MS).
However it is necessary to conduct further trials to find out the usefulness of these additives in case of other cultivars.
